What Is Emtricitabine-Rilpivirine-Tenofovir (Complera/Odefsey)?
Complera is a brand-name medication. The active ingredients in this product are emtricitabine, rilpivirine, and tenofovir disoproxil fumarate. Odefsey is also a brand-name medication. The active ingredients in this product are emtricitabine, rilpivirine, and tenofovir alafenamide.
Emtricitabine and tenofovir (both tenofovir disoproxil fumarate and tenofovir alafenamide) are nucleoside/nucleotide reverse transcriptase inhibitors (NRTIs). Rilpivirine belongs to a different class called non-nucleoside reverse transcriptase inhibitors (NNRTIs).
Complera is FDA-approved to treat HIV infection in adults and children weighing at least 35 kg. On the other hand, Odefsey is used to treat HIV infection in adults and children weighing at least 25 kg.
Complera and Odefsey do not cure HIV infection. However, when taken exactly as prescribed, they help control the virus, lower the amount of HIV in the blood (viral load), and reduce the risk of HIV-related complications.
Emtricitabine-Rilpivirine-Tenofovir Mechanism of Action
Emtricitabine and tenofovir block reverse transcriptase by acting as faulty building blocks during viral DNA synthesis, while rilpivirine binds directly to reverse transcriptase and inhibits its activity. Together, these medicines prevent HIV from making copies of itself, reducing the amount of virus in the blood (viral load) and helping protect the immune system.
Proper Use and Storage
- Take your doses exactly as directed. Take your medicine with a meal at approximately the same time each day.
- Don’t stop taking your medication without talking to your healthcare provider.
- If you miss a dose, take the missed dose immediately after you remember. Skip the missed dose if it is time for the next scheduled dose. Never take a double dose to compensate for the missed one.
- If you think an overdose has occurred, call the poison control helpline at 1-800-222-1222 immediately.
- Store Complera tablets in a tightly closed container at 25 °C (77 °F). Storage at temperatures between 15 and 30°C (59 to 86°F) is permitted for shorter periods, such as during transportation.
- Store Odefsey tablets in a tightly closed container below 30°C (86°F).
- Skipping doses or stopping treatment can allow HIV to become resistant to the medication, making it less effective.

Emtricitabine-Rilpivirine-Tenofovir (Complera/Odefsey) Dosing
Complera comes as purplish pink, capsule-shaped tablets to take by mouth. Each tablet contains:
- Emtricitabine 200 mg
- Rilpivirine 25 mg
- Tenofovir disoproxil fumarate 300 mg
Odefsey comes as gray, capsule-shaped tablets to take by mouth. Each tablet contains:
- Emtricitabine 200 mg
- Rilpivirine 25 mg
- Tenofovir alafenamide 25 mg
The recommended dose of Complera/Odefsey is one tablet once daily with food. Patients who become pregnant while virologically suppressed on Complera or Odefsey may continue therapy under the supervision of their healthcare provider with appropriate viral load monitoring.

Common effects include:
- Headache
- Difficulty falling asleep or staying asleep
- Diarrhea
- Nausea
- Tiredness
- Dizziness
- Depression
- Abnormal dreams
- Rash
Severe side effects include:
- Fever
- Skin blisters
- Mouth sores
- Redness or swelling of the eyes
- Swelling of the face, lips, mouth, or throat
- Trouble breathing or swallowing
- Pain on the right side of the stomach
- Dark “tea colored” urine
- Yellowing of the skin or eyes
- Light-colored stools
- Loss of appetite for several days
- Nausea
