What Is Bosutinib?
Bosutinib (boe-sue-ti-nib), available under the brand name Bosulif, is a targeted cancer medication used to treat chronic myeloid leukemia (CML), a type of blood cancer caused by an abnormal chromosome known as the Philadelphia chromosome.
Bosutinib belongs to a class of targeted cancer medications called tyrosine kinase inhibitors (TKIs). It works by blocking the abnormal BCR-ABL protein that promotes the growth of leukemia cells.
Bosutinib comes in the form of tablets or capsules and is taken orally (by mouth) with food. You may experience nausea, vomiting, diarrhea, or stomach pain during treatment with this medication.
Bosutinib (Bosulif): FDA-Approved Uses
The U.S. Food and Drug Administration (FDA) first approved Bosulif in 2012 for the treatment of Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML). Since then, its approved uses have expanded to include newly diagnosed chronic phase disease and certain pediatric patients.
Bosutinib (Bosulif) is FDA-approved to treat:
- Newly diagnosed chronic phase Philadelphia chromosome-positive chronic myeloid leukemia in adults.
- Chronic phase Ph+ CML that has not responded to or cannot tolerate previous therapies.
- Accelerated phase or blast phase Ph+ CML in adults who are resistant or intolerant to prior treatment.
- Chronic phase Ph+ CML in pediatric patients aged 1 year and older.
Important Safety Information
Bosulif may cause fertility problems in both female and male patients. This may affect your ability to have a child. Talk to your doctor if this is a concern for you.

Dosage
The dosage of bosutinib is determined based on the patient’s age, the phase of Ph+ CML (chronic, accelerated, or blast phase), and whether the condition is newly diagnosed or has been previously treated.
Bosutinib Usual Dosage for Adults
The recommended dose for newly diagnosed CML patients is 400 mg orally once daily.
For adults with previously treated chronic, accelerated, or blast phase Ph+ CML, the recommended dose is 500 mg taken orally once daily.
Bosutinib Usual Dosage for Children
The recommended dose of Bosulif for pediatric patients with newly diagnosed CP Ph+ CML is 300 mg/m2.
For children with chronic phase CML with resistance or intolerance to prior therapy, a dose of 400 mg/m2 is given orally once daily.
How It Is Taken
Bosutinib tablets and capsules are taken orally with food. Swallow the tablets or capsules whole. Do not crush, chew, split, or break them.
If swallowing capsules is difficult, the prescribed capsules may be opened and mixed with room-temperature applesauce or yogurt. Do not mix Bosulif with any other foods.
Caregivers should wear disposable gloves when preparing or administering the medication because contact with the contents of an opened capsule may be harmful.
Bosutinib Side Effects
Common side effects include:
- Diarrhea
- Headache
- Stomach pain
- Vomiting
- Nausea
- Rash
- Fatigue
- Liver dysfunction
- Fever
- Decreased appetite
- Respiratory tract infection
- Constipation
- Changes in certain blood tests
Serious side effects include:
- Severe diarrhea or other serious gastrointestinal problems (including severe stomach pain, persistent nausea or vomiting, or blood in the stool)
- Severe allergic reactions (symptoms may include trouble breathing, rash, stomach cramps, nausea, joint pain, fever, swollen face, lips, mouth, or tongue)
- Liver damage
- Fluid buildup (edema)
- Low blood cell counts, such as thrombocytopenia (low platelet levels), anemia (low red blood cells), and neutropenia (low neutrophil counts)
- Heart problems like heart failure or heart attack
- Kidney damage
