What Is Lumakras?

Lumakras (sotorasib) is a chemotherapy medication used for treating a specific type of non-small cell lung cancer (NSCLC). Providers prescribe Lumakras when NSCLC tumors meet the following conditions:

Surgical removal of the cancerous part of the lung(s) is not possible or the cancer has spread to other parts of the body (metastatic disease).
The tumor contains a specific gene called the KRAS-G12C gene, which is confirmed with testing.
One or more previous chemotherapy treatments have failed to stop the progression of the disease.

How Does Lumakras Work?

All human cells contain a gene that makes a protein called KRAS. This protein works to control and regulate the growth of cells.

If there’s the G12C mutation in the gene that makes KRAS, it results in the gene making the KRAS-G12C protein. This protein can result in cancer because cell growth is no longer properly controlled.

Among all people who have NSCLC, about 13% of people have tumors due to KRAS-G12C. Lumakras works by binding to KRAS-G12C and inactivating it so that it stops signaling cells to grow. In a clinical trial, one-third of the participants saw their tumors shrink by more than 30%. In addition, 2 of the 124 patients saw their tumors disappear.

What Is the Recommended Dose for Lumakras?

The typical dose of Lumakras is 960 mg (eight of the 120 mg tablets) by mouth once per day at the same time every day, with or without food. However, since Lumakras has a lot of stomach side effects, taking it with food might help. The medication is continued until your cancer has progressed or you’re no longer able to tolerate the medication. Your provider will adjust your dose down to 240 mg to 480 mg by mouth once per day if you’re experiencing certain side effects.

Swallow Lumakras tablets whole; don’t crush, chew, or split the tablets. If you’re having trouble swallowing tablets, you can place the tablets in 120 ml (4 ounces) of non-carbonated, room-temperature water without crushing the tablets. Don’t use any other liquid. Stir until the tablets are dispersed into small pieces (the tablets will not completely dissolve) and drink immediately or within 2 hours. Make sure to swallow the mixture and don’t chew the pieces of the tablet(s). Rinse the container with an additional 120 ml of water and drink.

People who miss their dose by more than 6 hours should skip that day’s dose and continue taking Lumakras the following day. Don’t double up and take an extra dose to make up for the missed dose. If you experience vomiting after swallowing the Lumakras tablet, don’t take another dose until the next day.
